Combating pancreatic cancer chemoresistance by triggering multiple cell death pathways

چکیده

Pancreatic cancer is the fourth most common cause of cancer-associated death in western countries, where incidence and number deaths are increasing every year. Intrinsic or acquired resistance tumor cells to chemotherapy agents major reason for failure traditional treatment. Several factors implicated this impressive resistance; however, these, it important highlight extensive cellular heterogeneity these tumors. This linked a wide range sensitivity that different clones same display chemotherapeutic agents. Accordingly, recent findings field have discovered new therapeutic targets order develop combinatory treatments, as well induce several cell pathways reduce therapy-threshold likelihood future resistance. research has focused on targeting mitochondria, an organelle with key roles regulating signaling pathways, such apoptosis, necroptosis, autophagy, ferroptosis, parthanatos. These — identifying compounds, alone combination, can target pancreatic ductal adenocarcinoma could be treatments.

Pancreatic Cancer Chemoresistance to Gemcitabine

Pancreatic ductal adenocarcinoma (PDAC), commonly referred to as pancreatic cancer, ranks among the leading causes of cancer-related deaths in the Western world due to disease presentation at an advanced stage, early metastasis and generally a very limited response to chemotherapy or radiotherapy. Gemcitabine remains a cornerstone of PDAC treatment in all stages of the disease despite suboptima...

Triggering of TLR7 and TLR8 expressed by human lung cancer cells induces cell survival and chemoresistance.

Compelling evidence suggests that inflammation, cell survival, and cancer are linked, with a central role played by NF-kappaB. Recent studies implicate some TLRs in tumor development based on their ability to facilitate tumor growth; however, to our knowledge, involvement of neither TLR7 nor TLR78 has yet been demonstrated. Here we have demonstrated expression of TLR7 and TLR8, the natural rece...

Multiple pathways of TWEAK-induced cell death.

TWEAK, a recently identified member of the TNF family, is expressed on IFN-gamma-stimulated monocytes and induces cell death in certain tumor cell lines. In this study, we characterized the TWEAK-induced cell death in several tumor cell lines that exhibited distinct features. Although the TWEAK-induced cell death in Kym-1 cells was indirectly mediated by TNF-alpha and was inhibited by cyclohexi...

Differential regulation of cell death pathways by the microenvironment correlates with chemoresistance and survival in leukaemia

Glucocorticoids (GCs) and topoisomerase II inhibitors are used to treat acute lymphoblastic leukaemia (ALL) as they induce death in lymphoid cells through the glucocorticoid receptor (GR) and p53 respectively. Mechanisms underlying ALL cell death and the contribution of the bone marrow microenvironment to drug response/resistance remain unclear. The role of the microenvironment and the identifi...

Stem cell pathways contribute to clinical chemoresistance in ovarian cancer.

PURPOSE Within heterogeneous tumors, subpopulations often labeled cancer stem cells (CSC) have been identified that have enhanced tumorigenicity and chemoresistance in ex vivo models. However, whether these populations are more capable of surviving chemotherapy in de novo tumors is unknown. EXPERIMENTAL DESIGN We examined 45 matched primary/recurrent tumor pairs of high-grade ovarian adenocar...
